Set in the fictional college town of Hilltowne, Charmed follows the lives of three sisters — Macy (Madeleine Mantock), Mel (Melonie Diaz), and Maggie Vera (Sarah Jeffery) — who, after the tragic death of their mother, discover they are three of the most powerful witches of all time. Together they possess the "Power of Three", which they use to vanquish supernatural demons and tear "down the patriarchy." Each sister has a unique magical power; the eldest Macy has the power of telekinesis, the middle sister Mel possesses the power of time-freezing, and the youngest Maggie has the power to hear people's thoughts/feel their emotions. After the season 3 finale, Maggie and Mel lose their power of three when their older sister Macy's dies however they discover from Guardian that there is another woman who restores power of three with them. 

Main characters:

• Madeleine Mantock as Macy Vaughn (1x01 - 3x18) - the Sister of Space and the late older sister of Mel and Maggie Vera. She is a "practical, driven, and brilliant geneticist." Macy initially moves to Hilltowne to work at the town's university lab, eventually discovering that she has two sisters. In 2021, she tragically died because of the Whispering Evil. Macy has the power of telekinesis, similar to the eldest sister Prue Halliwell in the original Charmed series.

• Melonie Diaz as Melanie "Mel" Vera (1x01 - 4x13) - the Sister of Time. She is "a passionate, outspoken activist", graduate student, and lesbian. Mel has the power of time-freezing, similar to the original middle sister Piper Halliwell in the original series.

• Sarah Jeffery as Margarita Emilia "Maggie" Vera (1x01 - 4x13) - the Sister of Emotion. She is "a bubbly college freshman" who is initially pledging into a sorority. Maggie has the power to hear people's thoughts, a change from the original youngest sister Phoebe Halliwell in the original series, who had the power of premonition.

• Lucy Barrett as Michaela "Kaela" Danso (4x01 - 4x13)  - a witch who will become a Charmed One after Macy Vaughn's death and, together with Mel and Maggie Vera, will have to restore the Power of Three. She is the late girlfriend of Dev. She has the power of Manifestation power due to Macy's stem cells who possess after Macy's death joins Mel and Maggie and restores the power of three, similar to the original fourth sister Paige Matthews in the original series. 

• Rupert Evans as Harry "Har" Greenwood (1x01 - 4x13) – a guardian angel who protects and guides witches. He is the late boyfriend of the eldest charmed one, Macy Vaughn who tragically died because of the Whispering Evil.

• Jordan Donica as Jordan "Jordy" Chase (2x01 - 4x13) - a law student, an amateur boxer, and he is also a sexy boxing instructor. He is the boyfriend of a younger charmed one, Maggie Vera.

• Ser'Darius Blain as Galvin Burdette (1x01 - 1x22) - Macy's late scientist colleague and ex-boyfriend.

• Ellen Tamaki as Niko Hamada (1x01 - 1x22) - Niko is a detective on the Hilltowne police force and Mel's ex-girlfriend.

• Valerie Cruz as Marisol Vera (1x01; Recurring: 3x12) - The girl's mother, a powerful witch.

• Natalie Hall as Lucy (1x01 - 1x20) - a sorority president and Maggie's former friend.

• Leah Lewis as Angela Wu (1x02 - 1x04) - a sexual assault victim who was possessed by the Harbinger of Hell.

• Brendon Zub as Trip Bailey (1x02 - 1x04) - Niko Hamada's detective ex-partner.

• Constantine Rousouli as Hunter Caine, a demonic assassin, Alistair's oldest son, Parker's older half-brother, and Abigael's oldest half-brother. He was involved with Maggie's mortal best friend, Lucy once.

• Craig Parker as Alistair Caine, benefactor of the Hilltowne's Department of Genetics and the father of Hunter, Abigael, and Parker.

• Rya Kihlstedt as Julia Wagner, Alistair's ex-wife and the human mother of Parker.

• Poppy Drayton as Abigael Jameson-Caine (2x03 - 3x17) - is a vengeful half-demon half-witch hybrid.

• Christin Park as Swan is an Asian woman who works at SafeSpace, Seattle.

• Eric Balfour as Julian Shea, a major investor in SafeSpace, Seattle.

• Shiva Kalaiselvan as Katrina Chandra, a practitioner of witchcraft and a medium.

• Shi Ne Nielson as Roxie, an owner of the magical nightclub "The Blue Camellia". She is also the eldest charmed one, Mel Vera's partner.

• Kapil Talwalkar as Dev was a singer in Roxie's nightclub before he was killed by Sunny. He was also the late boyfriend of a middle-charmed one, Kaela Danso.

• Natasha Henstridge as Diana is a new Whitelighter. She is from a different timeline.

In Season 5 Characters

• Julia Ormand as Marisa Reyes, a first cousin of Marisol Vera and a mother of Joséfina Reyes. She introduces herself to Maggie and Mel as 'Tía Marisa'.

• America Ferrera as Isadora Reyes, a second cousin of the Vera-Vaughn Charmed Ones; Macy, Mel and Maggie.
